British biotechnology startup Houdini Bio has officially launched out of stealth, securing approximately €1.7 million (£1.5 million) in pre-seed funding. The investment, led by SCVC, will advance the company's unique DNA sequence design platform. This innovative technology is engineered to overcome the cellular barriers that currently limit the effectiveness of genetic medicines.


Overcoming a Core Challenge in Gene Therapy

A fundamental hurdle holding back the full potential of gene and cell therapies is the body's natural defense system. Human cells often mistake therapeutic DNA for a viral invader and trigger mechanisms to "silence" the treatment. This premature shutdown restricts long-term effectiveness and can lead to disease progression or relapse for patients.

This cellular rejection, primarily driven by a master silencing mechanism known as the Human Silencing Hub (HUSH) complex, forces developers to use high, costly doses. This not only drives manufacturing costs but also increases the risk of dangerous immune reactions. The industry has long struggled with this challenge, often relying on unreliable trial-and-error screening methods.

A New Paradigm in DNA Engineering

Houdini Bio aims to solve this problem at the genetic level by introducing a shift to repeatable, engineering-driven DNA design. The company's platform combines machine learning with a deep mechanistic understanding of cellular silencing pathways. This allows for the re-engineering of therapeutic DNA, creating a form of genetic camouflage to evade the cell's defenses.

This novel approach preserves the medicine's intended function while making it invisible to the HUSH complex. The company was co-founded by CEO Jonathan Cohen-Gold and Chief Business Officer Lee Dunham, who bring complementary expertise to the venture. Cohen-Gold's PhD research at the University of Cambridge uncovered novel escape routes from the HUSH silencing mechanism. Dunham previously spent a decade at the Cell and Gene Therapy Catapult, witnessing firsthand the challenges faced by over 100 startups.

With backing from SCVC, Deep Science Ventures, and Cambridge Enterprise VC, Houdini Bio has demonstrated remarkable efficiency. The combined funding enabled the team to validate its technology and achieve its projected technical milestones in just six months. This rapid progress, accomplished in a third of the predicted time, underscores the platform's potential and the team's focused execution.

Investor Confidence and Market Positioning

Investors view Houdini Bio as a critical infrastructure provider for the next generation of medicine. Harry Destecroix, Managing Partner of lead investor SCVC, noted that the company solves a key biological bottleneck, transforming the efficacy and economics of gene therapies. He emphasized that the startup represents the exact type of deep-tech solution the ecosystem needs.
